DSIP: The Sleep Peptide Science Never Solved

The discovery
In 1974, Guido Schoenenberger and Marcel Monnier in Basel isolated a nonapeptide (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) from the cerebral venous blood of rabbits in whom sleep had been induced by electrical stimulation of the thalamus. Infused into recipient rabbits, the substance appeared to promote the slow-wave EEG activity of deep sleep – hence the name, delta sleep-inducing peptide.
The complication
Five decades on, DSIP is a scientific puzzle rather than a settled story. Replication of the sleep-EEG findings has been inconsistent across laboratories and models. No receptor for DSIP has ever been conclusively identified, and no gene encoding it has been found in genomic databases – an almost unique situation for a reportedly endogenous peptide, and one that has led some researchers to question what the original isolations actually captured. Meanwhile, scattered studies have attached it to other systems entirely: stress responses, luteinising hormone and growth hormone modulation, and pain models.
Why it is still studied
Precisely because the questions are open. A molecule with reported central effects, no known receptor, and no genomic origin is either an artefact worth ruling out or a doorway into biology nobody has mapped – and both possibilities are legitimate research programs. DSIP work today is a lesson in why negative and inconsistent results belong in the literature.
