Bacteriostatic Water vs Sterile Water for Peptide Reconstitution
Lyophilized peptides must be reconstituted before laboratory use, and the two standard diluents look identical on the shelf: bacteriostatic water and sterile water for injection. The difference is one ingredient, and it determines how long a reconstituted vial remains usable.
What each one is
Sterile water for injection (SWFI) is exactly that – water, sterilized, nothing added. Once a vial or a reconstituted solution is opened, nothing prevents microbial growth in it, so SWFI is a single-session diluent by design.
Bacteriostatic water is sterile water with 0.9% benzyl alcohol added as a preservative. The benzyl alcohol does not sterilize anything; it suppresses bacterial growth between uses. That is what makes multiple withdrawals from one vial over days or weeks a defensible practice – the pharmacopeial convention for multi-dose bacteriostatic vials is a 28-day working life after first puncture.
Which one suits which work
For a stock solution that will be sampled repeatedly across an experiment series, bacteriostatic water is the standard choice – each needle puncture introduces potential contamination, and the preservative is there to hold the line between sessions. For material that will be consumed in a single session, SWFI is sufficient.
The exception that matters: benzyl alcohol is cytotoxic at the concentrations used in cell culture work. If reconstituted peptide is destined for in-vitro cell-based assays, the benzyl alcohol carried into the medium can confound results or kill the culture outright. For that work, reconstitute in SWFI (or the assay’s specified buffer) and use the solution promptly, or dilute far enough that the preservative concentration becomes negligible – and note it in the method either way.
Practical handling either way
- Add diluent slowly down the vial wall and swirl gently – never shake. Foaming denatures and aggregates peptide.
- Refrigerate reconstituted solutions at 2-8 °C.
- Label every vial with the reconstitution date, concentration, and the lot number, so results stay traceable to that batch’s certificate of analysis.
