Lyophilization and Peptide Stability: Storage Practices for the Lab
Research peptides are almost always supplied as a lyophilized — freeze-dried — powder rather than in solution, and the reason is stability. This article covers why, and the storage and handling practices that preserve a peptide from the day it arrives to the end of a study.
Why peptides are lyophilized
Peptide bonds and side chains are vulnerable to hydrolysis, oxidation, and aggregation, and all three proceed far faster in solution than in the solid state. Lyophilization removes water under vacuum after freezing, leaving a stable cake or powder. Sealed under inert gas and kept cold, a lyophilized peptide can remain within specification for years; the same peptide in solution may degrade measurably in weeks.
Storing sealed vials
- Temperature: −20 °C is the standard recommendation for long-term storage of most lyophilized peptides. Short periods at ambient temperature — such as during shipping — are generally well tolerated by the dry powder.
- Light: store vials in the dark. Residues such as tryptophan, tyrosine, and methionine are susceptible to photo-oxidation.
- Moisture: lyophilized cakes are hygroscopic. Before opening a cold vial, allow it to reach room temperature so condensation doesn’t form on the powder — moisture uptake is one of the most common avoidable causes of degradation.
Reconstitution for laboratory work
For in-vitro work, peptides are typically reconstituted immediately before use. General practices from the analytical literature:
- Use an appropriate sterile diluent; bacteriostatic water (0.9% benzyl alcohol) is common in laboratory settings where a reconstituted stock will be sampled more than once, because it suppresses microbial growth.
- Add diluent slowly down the vial wall and swirl gently — vigorous shaking can cause foaming and aggregation, which reduces the usable concentration of intact peptide.
- Poorly soluble sequences may need a small volume of a secondary solvent (for acidic peptides, dilute base; for basic peptides, dilute acetic acid) before aqueous dilution — consult solubility data for the specific sequence.
After reconstitution
Reconstituted solutions should be refrigerated at 2–8 °C and used within the timeframe appropriate to the protocol; for longer holds, aliquoting and freezing single-use volumes avoids the repeated freeze–thaw cycles that degrade peptides in solution. Label every aliquot with the lot number so results remain traceable to the certificate of analysis for that batch.
