How to Reconstitute Lyophilized Peptides: A Laboratory Guide

Lyophilized peptides arrive as a dry cake because that is their most stable form. Reconstitution is where handling errors most often destroy material before an experiment starts.

Before you open the vial

Let a cold vial reach room temperature first. Lyophilized cakes are hygroscopic, and opening a chilled vial invites condensation directly onto the powder — the most common avoidable cause of degradation in otherwise well-stored material.

Choosing a diluent

Sterile water suits single-session preparations. Bacteriostatic water, containing 0.9% benzyl alcohol, suppresses microbial growth and suits stocks sampled repeatedly. Poorly soluble peptides may need a secondary solvent first — dilute acetic acid for basic sequences, dilute base for acidic ones.

The technique that matters

Add diluent slowly down the inside wall rather than onto the cake, then swirl gently until dissolved. Do not shake. Agitation foams the solution, and foaming drives aggregation, quietly reducing the intact peptide available to your assay.

Working out concentration

After reconstitution

Refrigerate at 2–8 °C and use within the window appropriate to your protocol. For longer holds, aliquot into single-use volumes and freeze — repeated freeze–thaw cycles degrade peptides faster than storage itself. Label aliquots with the lot number so results stay traceable to that batch’s certificate of analysis.
