CJC-1295 and ipamorelin appear together so often in research catalogues that they are easy to mistake for one compound. They are two unrelated peptides acting on two different receptors – and that difference is exactly why laboratories study them as a pair.
Two receptors, one axis
Growth hormone release from the pituitary is governed by complementary inputs: GHRH (growth-hormone-releasing hormone) stimulates somatotroph cells through the GHRH receptor, while ghrelin stimulates the same cells through a separate receptor, GHS-R1a. The two signals act through different second-messenger pathways and, in classic endocrinology experiments, their combination is synergistic rather than additive. Pairing a GHRH analogue with a ghrelin-receptor agonist recreates that two-input experiment with defined, synthetic tools.
CJC-1295: the GHRH analogue
CJC-1295 is built on the first 29 residues of human GHRH – the fragment that carries full receptor activity – with four amino-acid substitutions that protect it from rapid enzymatic cleavage. The name comes with an important fork: the version bearing a DAC group (Drug Affinity Complex – a maleimide linker that bonds covalently to serum albumin) persists in circulation for days, while the version without DAC, often called Mod GRF 1-29, retains a half-life of minutes. They are pharmacologically distinct tools: DAC produces a sustained elevation, no-DAC preserves pulsatile behaviour. Ipamorelin: the selective ghrelin-receptor agonist
Ipamorelin is a pentapeptide agonist of GHS-R1a, developed in the late 1990s. Its scientific claim to fame is selectivity: in the original characterisation work it stimulated growth-hormone release without the ACTH, cortisol, and prolactin activity seen with earlier compounds in its family (GHRP-2, GHRP-6). For receptor research, that clean profile is the point – effects observed in a model system can be attributed to the ghrelin receptor rather than a mix of off-target signals.

In research use, combining the two lets a model system probe the axis from both inputs at once – GHRH-receptor tone from the CJC-1295 side, ghrelin-receptor pulsing from the ipamorelin side.
